It contains Ser21-Leu431, which consists of two chains: Long chain A (Ser21-Phe177) and chain B (Ile179-Leu 431). The long chain A is further cleaved to yield a short chain A (Lys156-Phe 177) and N-terminal fragment (Ser21-Lys155). Molecular Weight The protein has a predicted MW of 47.5 kDa. Due to protein lysis and glycosylation, the protein migrates to 24-26 kDa(long chain A), 35-38 kDa(chain B) and 52-60 kDa(long chain A&chain B) based on Tris-Bis PAGE result. Physical Appearance Sterile Filtered White lyophilized (freeze-dried) powder. Purity > 95% as determined by SDS-PAGE and HPLC Bioactivity Measured by its ability to cleave a peptide substrate, N-carbobenzyloxy-Gly-Gly-Arg-7-amido- 4-methylcoumarin (Z-GGR-AMC). The specific activity is > 2000 pmol/min/g Endotoxin 1.0EU per 1g of the protein by the LAL method. Formulation Lyophilized from 0.22m filtered solution in 25mM HEPES, 150mM NaCl (pH 7.5). Normally 8% trehalose is added as protectant before lyophilization. Reconstitution Centrifuge tubes before opening. Reconstituting to a concentration more than 100 g/mL`is recommended.Dissolve the lyophilized protein in distilled water.
