FAQ COA 已发表文献 CD27, also known as TNFRSF7, is an approximately 55kDa transmembrane protein in the TNF receptor superfamily. It functions as a costimulatory molecule that supports lymphocyte activation and survival. Mature human CD27 consists of a 172aa extracellular domain (ECD) with three TNFR cysteinerich repeats, a 21aa transmembrane segment, and a 48aa cytoplasmic domain. Within the ECD, human CD27 shares 63%and 66%aa sequence identity with mouse and rat CD27, respectively. CD27 is expressed as a disulfidelinked homodimer that carries Nlinked and Olinked glycosylation. Proteolytic cleavage of CD27 results in the shedding of a 2832kDa fragment of the ECD. CD27 is weakly expressed on nave Tcells and NKcells and is upregulated upon cell activation. It is also upregulated on activated germinal center Bcells, plasma cells, and a subset of memory Bcells. CD27 binds to the transmembrane glycoprotein CD27 Ligand/CD70 which is expressed on activated Bcells, activated Tcells, and dendritic cells. This interaction contributes to the activation and survival of CD4+helper Tcells, CD8+effector Tcells, memory Tcells, and NKcells.
